ABSTRACT

Objective:To explore the effect of a Chinese medicinal composition ( Xiadanqi) on the prevention of radon exposure induced injuries of lung in vitro and in vivo. Methods:Mice were randomly divided into three groups of blank control group, radon-exposed group alone and radon-exposed group intervened with Chinese medicinal composition. The pathological changes of lung tissues in each group after 120 WLM were observed by HE and Masson staining, and the expressions of α-SMA protein and Vimentin protein in lung tissues were detected by immunohistochemistry staining. The levels of oxidative stress in lung tissue of each group were detected with SOD and MDA kits. At the same time, a radon exposed cell model and a radon exposure + Xiadanqi intervention cell model were constructed using an ecological radon chamber. The cell adhesion abilities of different groups were detected by an adhesion kit. The cell migration ability of each group was determined by the transwell migration experiment. The expression of E-cadherin and Vimentin protein was detected by Western blot. Results:Compared with the radon exposure group, the concentration of MDA was decreased ( t=4.43, P<0.05), the activity of SOD was increased ( t=3.22, P<0.05), and α-SMA and Vimentin protein expressions were decreased ( t=3.08, 7.57, P<0.05) in lung tissue of mice intervened with 2 mg/g Xiadanqi. In vitro, compared with radon exposure group, the migration ability was reduced ( t=4.78, 13.01, P<0.05), the cell adhesion property was enhanced ( t=3.41, 12.55, P<0.05), the expression of E-cadherin protein was increased ( t=2.96, 19.57, P<0.05), and the expression of Vimentin protein was obviously reduced ( t=21.00, 33.32, P<0.05) in radon-exposed cells with the treatment of Chinese medicine (150 μg/ml and 200 μg/ml). Conclusions:The Chinese medicinal composition ( Xiadanqi) has a certain radioprotective effect on radon exposure induced injury by reducing oxidative stress, attenuating EMT and fibrosis, and thus it may be applied as a protective agent for radon induced injury.

ABSTRACT

BACKGROUND: Human leukocyte antigen-G (HLA-G) is a tumor-associated molecule, whose expression may help the cancer cells to escape the immune response. AIMS: The aim of this study was to evaluate the diagnostic value of HLA-G level in oral squamous cell carcinoma (OSCC). MATERIALS AND METHODS: A total of 52 patients who had definite pathological diagnosis and 20 cases of healthy controls were enrolled in this clinical trial. Immunohistochemisty (IHC) and quantitative real-time reverse transcription-polymerase chain reaction (RT-PCR) analysis were considered for HLA-G identification and multilevel validations. Statistical analysis was performed using SPSS and statistical significance was determined at P < 0.05. RESULTS: IHC results demonstrated that the expression of HLA-G in OSCC was strongly positive and the rate of positive expression was 55.77% (29/52), but the expression of HLA-G in healthy controls was negative (0/20). Furthermore, RT-PCR results showed that the positive expression rate of HLA-G messenger RNA was weak in healthy controls, but strong in OSCC. Besides, HLA-G expression in the tumors was significantly correlated with histological grade. CONCLUSIONS: Our results suggested that HLA-G is associated with the prognosis of OSCC and may serve as a novel therapeutic target.

ABSTRACT

Objective To observe the effect of Yunnanbaiyao combined with gelatin sponge on wound healing in patients with post-extraction hemorrhage. Methods 72 patients received dental extraction were selected, and they were divided into observation group (Yunnanbaiyao + gelatin sponge) and control group (gelatin) according to the digital table, each group in 36cases.The hemostasis after treatment and the improvement of pain after 12h and 24h after treatment were observed. The wound healing was observed in the two groups after 7 days of treatment. Results After treatment, the total effective rate was 91.67% in the observation group and 69.44% in the control group. The effective rate of hemostasis in the observation group was significantly higher than that of the control group(x2 =7.32,P =0.007).The VAS scores of the observation group at 12 h (t =23.44,P =0.000) and 24h (t =22.86,P =0.000) after pack treatment obviously decreased; The VAS scores of the control group at 12h(t =19.87,P =0.000) and 24h (t =18.47,P =0.000) after pack treatment obviously decreased; The VAS scores of the observation group at 12h and 24h after pack treatment was lower than those of the control group, the differences were statistically significant(t =7.03,5.03,all P =0.000).The wound healing of the observation group was improved after 7d treatment(t =8.12,P =0.00);The wound healing of the control group was improved after 7d treatment(t =5.39,P =0.00);The wound healing of the observation group was better than that of the control group, the differences were statistically significant (t =2.88,P =0.005).Conclusion Yunnanbaiyao combined with gelatin sponge can relieve the bleeding symptoms after tooth extraction, promote wound healing, reduce the patients' pain.

ABSTRACT

Objective:To study the relation among nm23-H 1 expression,DNA contents and lymph node metastases of head and neck squamous cell carcinoma(HNSCC).Methods:Fresh specimens from 30 previously untreated patients with HNSCC were examined for the determination of nm23-H 1 expression with immunohistochemistry, Aneuploid and S-phase fraction(SPF) with flow cytometry(FCM).The association among these parameters with lymph node metastases was studied. Results: nm23-H 1 expression was found in 18 cases (60%).and aneuploid in 20 (66.7%).Correlation was found between lymph node metastasis and nm23-H 1 protein expression (P
